Sections from a 3D image of histoplasma capsulatum yeast cells provided by Dr. William Goldman (Department of Molecular Microbiology, Washington University School of Medicine, St. Louis). The raw image was collected with a 100x / 1.35 NA oil-immersion objective lens at a fluorescent wavelength of 520 nm. Pixel sizes are 0.1 micrometers laterally and 0.15 micrometers axially. The 3D image was processed with three XCOSM algorithms: LLS, EM, and EM with intensity penalty (EM-IP) The LLS image was generated using a processing parameter of 0.01. The EM image is the result after 1000 iterations of the EM algorithm. The EM-IP image is the result after 1000 iterations of the regularized EM with intensity penalty using a parameter of 1e-6.
